Propagation based phase contrast x-ray imaging of Caenorhabditis elegans

چکیده
Introduction: Imaging of multicellular organisms with nano-scale resolution is one of the challenges of modern life sciences. To solve biological problems, often the combined information of several microscopy techniques is necessary. While confocal fluorescence microscopy and optical super-resolution fluorescence can detect the distribution of markers in live cells, electron microscopy (EM) offers highest resolution in fixed specimen. This, at the price of invasive sample preparation involving fixation, cutting and staining. Yet another important contrast is provided by x-ray microscopy which can yield quantitative density distribution in bacterial cells[1]. At the P10 nanofocus holography endstation at PETRAIII we performed a full-field phase contrast x-ray tomography on the multicellular nematodes Caenorhabdits elegans which serve as model organism for biological research fields including neuroscience, cell biology and signaling[2, 3]. This correlative imaging experiment uses protocols from EM[4] and molecular neurogenetics [5].

Propagation-based x-ray phase-contrast imaging with broad focus conventional x-ray sources
A propagation-based x-ray phase-contrast imaging (PBI) setup using a conventional x-ray source (LFF Cu target) is presented. A virtual x-ray source of 40 x 50 μm was created by using, horizontally, a 6 take-off angle (with the x-ray tube working in the line focus geometry) and, vertically, a 50 μm slit . The sample was set 12 m from the source. Propagation-based x-ray phase-contrast (PB) image ...
متن کاملComparison of phase contrast X-ray computed tomography methods for non-destructive testing of materials
Currently the basis for standard X-ray computed tomography (CT) is absorption. A volumetric map of a specimen in three dimensions is generated from a set of absorption radiographs. The contrast of details strongly depends on a difference in absorption coefficient between the detail and the environment. However, when the absorption difference is low, sufficient contrast for a good quality X-ray ...
متن کاملComputational techniques in propagation–based x–ray phase imaging
X–ray phase imaging utilizes a variety of techniques to render phase information as intensity contrast and these intensity images can in some cases be processed to retrieve quantitative phase. A subset of these techniques use free space propagation to generate phase contrast and phase can be recovered by inverting differential equations governing propagation. Two techniques to generate quantita...

A full-field hard X-ray imaging beamline (BL-4) was designed, developed, installed and commissioned recently at the Indus-2 synchrotron radiation source at RRCAT, Indore, India. The bending-magnet beamline is operated in monochromatic and white beam mode. A variety of imaging techniques are implemented such as high-resolution radiography, propagation- and analyzer-based phase contrast imaging, ...
متن کاملA feasibility study of X-ray phase-contrast mammographic tomography at the Imaging and Medical beamline of the Australian Synchrotron.
Results are presented of a recent experiment at the Imaging and Medical beamline of the Australian Synchrotron intended to contribute to the implementation of low-dose high-sensitivity three-dimensional mammographic phase-contrast imaging, initially at synchrotrons and subsequently in hospitals and medical imaging clinics. The effect of such imaging parameters as X-ray energy, source size, dete...
